“指示” means an order or an instruction which your boss or senior gives you. Basically, “指示” is used as business situations and the way to use it is only two situations. When you give an order, you can say “指示をする”, however, native speakers often omit を, so they often say “指示する” instead of “指示をする”. Or when you are given an order by someone, you can say “指示をされる”, and also they often omit を, so they often say “指示される” instead of “指示をされる”. When native speakers use “指示する” or “指示される” in daily conversations, it is sometimes said “言う” or “言われる” instead of “指示する” and “指示される”.
